When a search engine looks at your website it scans for words and phrases input by a person doing a search query. The density of certain keywords, related to the initial search query, is evaluated for relevance to the search terms used. In short they read your webpages and look for keyword matches.
As a webmaster your task is to find out what search terms (i.e. keywords) you want your prospective visitors to use to find your website.
